Preformulation Considerations Development and Evaluation of Mesalamine Loaded Polysaccharide-Based Complex Mucoadhesive Beads for Colon Targeting

Abstract: Background: Inflammatory bowel disease (IBD) is a chronic inflammatory of large intestine consisting ulcerative colitis and Crohn’s disease. Mesalamine first line drug use for the treatment but it has drawback low water solubility permeability. In addition, oral delivery to colon faces problem gastric degradation thus increases dose size frequency. Objectives: The present study was aimed assay mesalamine develop an enteric coated loaded mucoadhesive beads using guar gum, sodium alginate carbopol 940 effective colon. Methods: Preformulation done check purity drug. were prepared by ion gelation methods with Eudragit S100 characterized. Results: melting point absorbance maxima found be 282°C 220 nm respectively. FTIR confirms polymers compatible. A total 5 different formulations which specifically release in region sustain fashion. formulation MA-F4 chosen as optimized formulation. Particle size, Zeta potential, %EE, %Yield, %DL vitro 445.6 ± 67.1 μm, -28.01 0.16 mV, 94.98 3.22, 96.27 30.4 0.9 95.07 3.85 Conclusion: demonstrated that can sustained manner helps management IBD.
